IDK, they can't even chart on Bubbling Under with a Korean song(cmiiw) so I think it's going to be ALOT harder for them to chart with an English song. It'd be a win for them to chart there though considering it's not very easy as KPOP stans think it is to chart on the Hot100. And are they getting radioplay? That might help their chances alot.
Either way their international growth has been really impressive, perched for their next full album.

I guess so, but them not being able to chart in Bubbling Under with a Korean song shows that they just don't have that many people checking for them in the US. Their fanbase there, at least compared to BTS\BP, is pretty small.
BLACKPINK were able to chart in BU before they started regularly charting on Hot100, same for BTS.
Anything is possible and they could go straight to charting on the Hot100, but I haven't seen it before.

If I am not mistaken there was a thread about them doing good on amazon, it's already promising. How did they do on US youtube main chart page? How did they do on US spotify, apple music and itunes? If they got top5 or close everywhere then why not? Do they have radioplay? To chart a single on hot100 or bubbling under is not the same as to do it for a title of whole album, it should be easier cause fans are totally focused on 1 song. I have a feeling they can do it, at least on 90-100 position. Have you checked any predictions?

well right now they only have uk 100 singles and rolling stone plus radio plays

Difficult to say Republic did an almost non existent job promoting the song. Poor playlisting, sending broken links to fans, asking us would we like to hear it on radio after the tracking ended....just a mess.
It's hard in todays day and age when most of the chart is songs pushed to over 1k Spotify playlist to compete if your own label doesn't play that game. JYPE basically left it in the fans hands and expected them to do 90% of the work while other companies pay for playlisting and radio spin deals.
it's an easy entry into hot 100 imo for twice. Twice have already got enough digital sales to enter and fans did their job purchasing.
